PEG-MGF is a pegylated form of mechano growth factor (MGF), a peptide derived from a splice variant of insulin-like growth factor 1 (IGF-1). The attached polyethylene glycol (PEG) is intended to slow clearance and enzymatic breakdown by increasing the molecule’s size. It is supplied in 2mg lyophilized vials.
The IGF-1 gene produces several isoforms through alternative splicing of its E domain. MGF corresponds to the isoform called IGF-1Ec in humans and IGF-1Eb in rodents, and it carries a unique C-terminal E-domain peptide. Geoffrey Goldspink’s group in London named it mechano growth factor after finding that its expression rises in skeletal muscle subjected to stretch or mechanical loading.
Research on the MGF E peptide has examined activation and proliferation of muscle satellite cells, often reporting that it behaves differently from mature IGF-1, which is more closely tied to differentiation. Other studies have looked at cardiomyocyte and neuronal survival in injury models. Native MGF peptide is unstable in serum and has a very short half-life. PEGylation is a standard technique for extending the circulation time of peptides and proteins, so the pegylated version allows longer experimental windows than the native sequence.
