Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. It is the unmodified parent compound from which the acetylated and amidated Semax analogs in this catalog are derived. Structurally, it is an analog of the ACTH(4-10) region of adrenocorticotropic hormone.
Researchers at the Institute of Molecular Genetics of the Russian Academy of Sciences created Semax by fusing the ACTH(4-7) tetrapeptide to Pro-Gly-Pro. The ACTH portion was chosen for its reported activity in the nervous system, while the added tripeptide slows breakdown by peptidases. The resulting peptide lacks the adrenal-stimulating activity of full-length ACTH.
Most published data come from rodent experiments. Studies have reported increases in BDNF and TrkB expression in the hippocampus, changes in genes involved in immune and vascular function after cerebral ischemia, and effects on dopamine and serotonin turnover. Because Semax has free termini, it serves as the reference compound when laboratories measure how N-acetylation or C-terminal amidation alter peptide stability.
