CJC-1295 without DAC, also called modified GRF (1-29) or Mod GRF 1-29, is a 29-amino-acid analogue of growth hormone-releasing hormone (GHRH). It is the GHRH(1-29) fragment with four amino acid substitutions that make it more resistant to enzymatic breakdown, and it lacks the drug affinity complex (DAC) that gives CJC-1295 with DAC its long half-life.

What is CJC-1295 without DAC?

Native GHRH(1-29) is cleaved within minutes by dipeptidyl peptidase-4 (DPP-4) at the Ala2 position. Mod GRF 1-29 replaces four residues (D-Ala2, Gln8, Ala15 and Leu27) to slow this cleavage and reduce oxidation and rearrangement. Like native GHRH, it binds the GHRH receptor on pituitary somatotroph cells and raises cAMP, which triggers growth hormone release. Because it has no albumin-binding group, its action is short and pulse-like, in contrast to the sustained elevation reported for the DAC version.

It is often studied together with ghrelin-receptor (GHS-R1a) agonists such as ipamorelin, because the two receptor pathways amplify growth hormone release in pituitary models. That pairing is available as the CJC-1295 / Ipamorelin blend.
