Description
Larazotide acetate (AT-1001) is a synthetic octapeptide with the sequence Gly-Gly-Val-Leu-Val-Gln-Pro-Gly. Its design was based on a fragment of zonula occludens toxin (Zot), a protein produced by Vibrio cholerae. The compound is studied as a regulator of epithelial tight junctions. Its short length and water solubility make it simple to add to culture media at defined concentrations.
What is Larazotide?
Tight junctions are protein complexes that seal the space between neighboring epithelial cells and control paracellular permeability. Zonulin, a protein related to Zot, has been proposed to loosen these junctions, and larazotide was developed to oppose this signal. In vitro studies with intestinal epithelial cell lines such as Caco-2 have measured its effect on transepithelial electrical resistance (TEER) and on the redistribution of junctional proteins like ZO-1 and actin after exposure to gliadin fragments or inflammatory stimuli.
The peptide has also been evaluated in clinical research programs related to celiac disease, which drew wider attention to the tight junction as a research target. In the laboratory it provides a defined tool for probing paracellular transport, cytoskeletal rearrangement, and barrier function in cell monolayers and animal models.
