Description

Nicotinamide adenine dinucleotide (NAD+) is a coenzyme found in every living cell. It consists of two nucleotides, one carrying adenine and one carrying nicotinamide, joined through their phosphate groups. It is a small molecule, not a peptide.

What is NAD+?

NAD+ has two broad roles. As a redox cofactor it accepts electrons in glycolysis, the TCA cycle and fatty acid oxidation, cycling between NAD+ and NADH. As a consumed substrate it is broken down by sirtuins (protein deacylases), poly(ADP-ribose) polymerases (PARPs) involved in DNA repair, and the NADase CD38. Cells replace it through salvage pathways from nicotinamide, nicotinamide riboside (NR) and nicotinamide mononucleotide (NMN). Rodent studies have reported lower NAD+ levels in several tissues with age, which is why the NAD+ metabolome is a common target in ageing research.
