Description
Retatrutide (LY3437943) is a 39-amino-acid synthetic peptide that activates three receptors: GLP-1, GIP and glucagon. It is one of the first “triple agonists” to reach late-stage clinical trials and one of the most closely followed compounds in metabolic research.
What is Retatrutide?
Retatrutide was developed as a successor to dual GLP-1/GIP agonists such as tirzepatide. Adding glucagon receptor activity is the feature researchers study most, because glucagon signalling is linked to hepatic glucose output and energy expenditure in animal models.
- GLP-1 receptor: glucose-dependent insulin secretion and satiety signalling
- GIP receptor: incretin signalling and adipose tissue metabolism
- Glucagon receptor: hepatic glucose output, lipid metabolism and energy expenditure
Research Applications
- Obesity and energy-balance models
- Insulin resistance and glucose homeostasis
- Hepatic lipid metabolism and fatty-liver models
- Receptor pharmacology comparisons with mono- and dual-agonists
